Human iPS cells have a great potential to be cell sources for regenerative medicine because of the promise of infinite self-renewal and the capability to differentiate into multiple cell types. This book focuses on another great potential of human iPS cells, which is the establishment of human disease models using patient-specific iPS cells. Human iPS cells can be easily obtained from a patient’s somatic cells and provide the entire information on the patient’s genome. Accordingly, we can generate disease models for inheritable diseases in cell culture dishes using iPS cells. This is a quite new technique but holds tremendous potential for our increased understanding of pathogenesis, and will then be the basis for novel drug development industries. All the authors are leading researchers in this field and they have reported many kinds of patient-derived iPS cells. In this book, they introduce the aspects that could be recapitulated in terms of disease modelling as well as further innovative findings such as novel pathogenetic insights and novel therapies.

Tetramer Stability and Functional Regulation of Tumor Suppressor Protein p53

This thesis presents the first report of the comprehensive and quantitative analysis of the effects of tumor-derived mutations on the tetrameric structure of tumor suppressor protein p53, which plays a central role in maintaining genomic integrity. Inactivation of p53 via mutation of its gene is a key step in tumorigenesis. Biophysical analyses revealed that the stability of the mutant peptides varied widely. Formation of a tetrameric structure is to be critical for protein–protein interactions, DNA binding, and the post-translational modification of p53. A small destabilization of the tetrameric structure therefore could result in dysfunction of tumor suppressor activity. This work suggests that the threshold for loss of tumor suppressor activity, in terms of the disruption of p53’s tetrameric structure, could be extremely low. Furthermore, functional control of p53 via tetramer formation was demonstrated, based on the structure–function analysis of mutant p53. The results disclosed that relatively small changes in tetramer formation, induced by the stabilization or inhibition of homo-tetramerization, could control p53 function.

However, in the 1990s, reactive oxygen species have been recognized to be involved in reversible structural alterations of molecules: The homeostatic modulation of oxidant levels is a highly efficient mechanism that allows all cells to tightly control their redox status within a very narrow range. Especially the activity of the Rel/NF-kB family of transcriptional factors that regulate responses in inflammation, reperfusion injury and apoptosis is controlled by intracellular reactive oxygen species levels. Our understanding of how these transcriptional factors are negatively or positively regulated by oxidative stress has since greatly increased. Questions for future investigation center on the modulation of oxidative stress by personal genome information for the prevention of several pathologic states.
